The 'hosts' file from your computer, right?

The easy way:
If you have Tiny Umbrella on that computer, just go to Advanced tab (the right side of TU's window) and untick 'Set Hosts to Cydia on Exit'.

The geeky way:
1) Browse to Start -> All Programs -> Accessories
2) Right click “Notepad” and select “Run as administrator”
3) Click “Continue” on the UAC prompt
4) Click File -> Open
5) Browse to “C:\Windows\System32\Drivers\etc”
6) Change the file filter drop down box from “Text Documents (*.txt)” to “All Files (*.*)”
7) Select “hosts” and click “Open”

8) Add '#' in front of '74.208.10.249 gs.apple.com' and close Notepad. Save when prompted.

You might want to undo the changes once you are unlocked. Just delete the '#' or tick 'Set Hosts to Cydia...'

You don't need to install any program to remove the cydia entry. Just open it up with notepad/wordpad, edit it and save it.

That's it.
